# Clinical Daily Summary: What Happened on October 6, 2026

**Survodutide cuts weight by up to 13.1% in Phase 3 obesity trial**

Epcoritamab lowered progression-or-death risk in lymphoma. Other updates covered asthma, pregnancy-risk prediction and multiple myeloma.

## Today in brief

Boehringer Ingelheim’s survodutide met both primary endpoints in a 76-week Phase 3 trial in adults with obesity or overweight and type 2 diabetes, with up to 13.1% weight loss versus 3.1% on placebo. [1] [2] Gastrointestinal side effects led 18% of participants on survodutide to stop treatment, compared with 1.2% on placebo. [2]

Genmab and AbbVie’s Phase 3 trial found that adding epcoritamab to R-CHOP lowered the risk of progression or death by 51% in newly diagnosed diffuse large B-cell lymphoma. [3] [4] The combination remains investigational for this use, and the companies plan to present full results at a future medical meeting. [3]

## Survodutide’s weight loss came with frequent treatment stops

The 755-person SYNCHRONIZE-2 trial compared weekly survodutide at two doses with placebo for 76 weeks in adults with overweight or obesity and type 2 diabetes. [2] In an analysis assuming participants stayed on treatment, average weight loss reached 13.1%, versus 3.1% with placebo, and up to 79.3% lost at least 5% of their weight, versus 32.7%. [2] When the analysis included outcomes regardless of treatment discontinuation, weight loss was 9.8% with the higher dose and 8.2% with the lower dose, compared with 3.9% on placebo. [2]

Survodutide lowered hemoglobin A1c, a measure of average blood sugar, by up to 1.21 percentage points from a baseline of 7.4%, compared with 0.03 points on placebo. [2] Gastrointestinal events led 18% of survodutide recipients to stop treatment, compared with 1.2% of placebo recipients, usually during dose escalation. [2] Boehringer Ingelheim said a fixed dose-escalation schedule may have contributed to the treatment stops. Newer trials allow more flexible dose increases, the company said. [2]

The trial did not compare survodutide with an approved incretin drug, so its performance against those treatments remains unknown. [2] Survodutide is not approved, and the companies expect cardiovascular outcomes data from SYNCHRONIZE-CVOT later in 2026. [1] [2]

## Epcoritamab combination cuts lymphoma progression-or-death risk

An interim analysis of EPCORE DLBCL-2 found that epcoritamab plus R-CHOP reduced the risk of disease progression or death by 51% compared with R-CHOP alone. [3] [4] The combination significantly improved progression-free survival, the time patients live without their disease worsening or dying. [3] The open-label trial compares the combination with R-CHOP alone, not with Polivy plus R-CHOP. [3] [4]

The Independent Data Monitoring Committee recommended unblinding the study after observing the benefit. [3] The companies said the safety profile was generally consistent with prior experience of the medicines. [3] Epcoritamab is authorized in the European Union for some patients whose diffuse large B-cell lymphoma has returned or resisted at least two treatment regimens, but its use with R-CHOP for newly diagnosed disease remains unapproved. [3] [4] AbbVie and Genmab plan to present complete results at a future medical meeting and discuss next steps with regulators. [3]

## Also worth knowing

- Vial dosed the first participant in a Phase Ib asthma study of its bispecific antibody, which blocks IL-13 and TSLP. The trial will assess safety, tolerability, lung function and inflammation markers. [5]
- A retinal-imaging AI platform predicted preeclampsia with an AUC of 0.81 in an independent cohort of 79 pregnancies, without retraining after development in 1,267 pregnancies. [6]
- In follow-up of four people with relapsed or refractory multiple myeloma treated with in vivo BCMA CAR-T, all initially responded, but three relapsed within five months and later died. Median progression-free survival was four months. [7]
- On October 5, the FDA approved Alvotech’s second Reykjavik production suite for SIMLANDI, doubling capacity available for US supply. [8]
- On October 5, Novo Nordisk said the FDA extended its review of denecimig for hemophilia A while manufacturing-facility remediation continues. The company is targeting a possible US launch in the first half of 2027. [8]
- On October 5, AMETHYST data showed skin improvements through 52 weeks with litifilimab in cutaneous lupus erythematosus. Phase 3 results are expected in the first half of 2027. [8]

## The day in numbers

- **13.1% vs. 3.1%**: Average weight loss with survodutide and placebo in the analysis assuming participants remained on treatment. [2]
- **18% vs. 1.2%**: Participants who stopped survodutide or placebo because of gastrointestinal events. [2]
- **51%**: Lower risk of progression or death with epcoritamab plus R-CHOP versus R-CHOP alone in an interim lymphoma analysis. [3]
