# Clinical Daily Summary: What Happened on November 2, 2026

**Povorcitinib hits the primary endpoint in two vitiligo trials**

Tezspire improved symptoms and tissue inflammation in a Phase 3 trial. Other findings cover ALS, wearable alerts for hypertensive crises and midlife hormone therapy.

## Today in brief

Povorcitinib met the primary endpoint in two Phase 3 trials, with more patients achieving substantial facial repigmentation than on placebo. [1] In a separate Phase 3 trial, Tezspire improved swallowing symptoms and tissue inflammation in eosinophilic esophagitis, with benefits lasting through 52 weeks. [2]

Shanghai Zhimeng Biopharma completed enrollment in a 255-patient Phase 2/3 ALS trial, while researchers presented wearable-based predictions of hypertensive crises up to 72 hours ahead. [3] [4] A long-term observational study found no apparent increase in cardiovascular events among women who began hormone therapy in midlife. [5]

## Povorcitinib met the primary endpoint in both vitiligo trials

Both placebo-controlled Phase 3 STOP-V trials met their week 52 primary endpoint: at least a 75% improvement in facial Vitiligo Area Scoring Index. [1] In STOP-V1, 18.9% of patients taking povorcitinib reached that threshold, compared with 6.8% on placebo. In STOP-V2, the rates were 18.9% and 3.1%. [1]

STOP-V1 enrolled 467 patients and STOP-V2 enrolled 449. The proportion experiencing any treatment-emergent adverse event was similar between drug and placebo groups in both trials. [1] The study authors said the findings support povorcitinib as an effective, well-tolerated systemic treatment for adults with moderate-to-severe nonsegmental vitiligo. [1]

## Tezspire improved EoE symptoms and tissue inflammation

Tezspire improved tissue inflammation and swallowing symptoms in the Phase 3 CROSSING trial in eosinophilic esophagitis, AstraZeneca and Amgen said. [2] The benefits lasted through 52 weeks. [2]

## Also worth knowing

- Shanghai Zhimeng Biopharma enrolled 255 patients at 15 mainland China centers in its Phase 2/3 CB03-154 ALS trial. The primary measure is change in ALSFRS-R score at week 39. The company expects top-line Phase 2 results in the third quarter of 2027 and says it plans to seek conditional approval in China if the results are positive. [3]
- A study tracked 4,190 adults with stage 2 hypertension in Pakistan and found wearable data analyzed across 47 features may predict a crisis within 72 hours. When doctors received alerts and changed medication, crises fell 61% and emergency visits fell 45%. The single-hospital findings need further study before they can guide wider care. [4]
- In an observational study, more than 52,000 women were followed for an average of 12 years. Midlife hormone therapy was not associated with higher adjusted risk of cardiovascular disease, blood clots or premature death, though differences between women who chose treatment and those who did not could affect the results. [5]

## The day in numbers

- **18.9%**: Share of patients on povorcitinib who reached the primary endpoint in each vitiligo trial, compared with 6.8% and 3.1% on placebo. [1]
- **52 weeks**: How long benefits persisted in the Phase 3 eosinophilic esophagitis trial of Tezspire. [2]
- **255**: Patients enrolled in Zhimeng's Phase 2/3 ALS trial. [3]
