# Research Daily Summary: What Happened on October 28, 2026

**Immune-cell maps link genetic variation to risk across 16 diseases**

Researchers also found an early pancreatic-cancer signal in CT scans, tested an H5N1 vaccine in cows and built a tool to map gene regulation.

## Today in brief

Three studies measured genetic activity in 5.4 million immune cells from 1,925 Australians, linking immune-cell involvement to risk regions for 16 diseases and nearly 1,500 potentially involved genes. [1] Separately, Mayo Clinic’s REDMOD system detected pancreatic-cancer signals in scans an average of 475 days before diagnosis, while vaccinated cows had about 1,000-fold lower peak H5N1 viral RNA in milk after challenge. [2] [3]

## Immune-cell studies link genetic variation to disease risk

Researchers measured genetic activity in 5.4 million immune cells across 28 cell types from 1,925 Australians. [1] Across three studies, they linked immune-cell involvement to genetic risk regions for 16 diseases and identified nearly 1,500 genes that may contribute to disease. [1]

Chromatin accessibility, a measure of how open DNA is to regulatory activity, may explain up to 22.6% more disease-linked genetic regions than gene activity alone. [1] The researchers also found a variant that raises OSM expression in natural killer cells and is linked to greater irritable bowel disease risk. [1]

## AI detected pancreatic-cancer signals before diagnosis

In a retrospective study, Mayo Clinic’s REDMOD system detected pancreatic-cancer signals in CT scans an average of 475 days before diagnosis. [2] It was nearly twice as sensitive as experienced radiologists in an evaluation of 219 patients whose scans had not been identified as showing cancer when taken. [2]

Among 539 people without pancreatic cancer, REDMOD classified just over 81% of scans as cancer-free. It correctly classified 87.5% in an 80-person NIH-PCT dataset. [2] The team said further research, including studies in high-risk patients and prospective, multicenter validation, is needed before routine clinical use. [2]

## An mRNA vaccine reduced H5N1 viral RNA in cows’ milk

Four lactating cows received two doses of an mRNA–lipid nanoparticle vaccine encoding H5 hemagglutinin, while four control cows received saline. [3] After all eight cows received an H5N1 challenge, vaccinated animals had about 1,000-fold lower peak viral RNA in milk, along with less mastitis and mammary fibrosis. [3]

Researchers detected no vaccine-derived H5 mRNA in milk samples collected during the eight days after vaccination and reported no obvious adverse effects. [3] The experiment involved eight cows, challenged 49 days after the first dose and monitored for 14 days after infection. [3]

## FIRE maps gene-regulatory differences across the genome

University of Utah researchers developed FIRE, which combines machine learning and long-read DNA sequencing to find regulatory regions and compare activity between maternally and paternally inherited chromosome copies. [4] FIRE identified tens of thousands of previously overlooked regions more likely than other regions to contain disease-associated variants. [4]

In a celiac-disease-associated variant, changing one DNA letter altered the accessibility of a nearby gene switch in laboratory experiments. [4] Most newly identified regulatory differences have unknown biological functions. [4]

## Also worth knowing

- An engineered Cas12m system delivered up to eight guide RNAs in one AAV vector to activate six genes in mice, with phenotypic rescue in models of Duchenne muscular dystrophy and Scn1a haploinsufficiency. [5]
- In mice, engineered albumin extended erythropoietin’s plasma half-life to 5.7 days, compared with 1.1 days with standard albumin, and delivered five times more fusion protein into blood after nasal administration. [6]
- In preclinical psoriasis models, antibodies against CHI3L2 reduced inflammatory responses, keratinocyte proliferation and skin inflammation. [7]
- Experiments in mice and laboratory studies found that MCT1-mediated lactate transport into mitochondria helps the heart adapt to stress. Disrupting the transporter impaired that response in stressed mice. [8]
- MIT researchers’ BIOSENTER robot traveled about two meters through collapsed small intestine in live pigs in 11 minutes, performing imaging, tissue sampling and fluid delivery without visible tissue injury. [9]
- A three-photon microscopy platform recorded neural activity at single-cell resolution about 2 millimeters deep in living mouse brains, compared with about 1.2 millimeters for earlier functional recordings. [10]
- A systematic study mapped how short sequences in the 3′ untranslated region affect RNA abundance and localization, finding a broadly additive regulatory pattern. [11]
- A review described genome mining, metagenomics and cultivation devices as ways to find compounds from underexplored microbes. Only 11% to 32% of large-scale attempts to express natural-product pathways in alternative hosts yielded detectable compounds. [12]
- Researchers found that inhibitor of apoptosis proteins help some chemotherapy-treated cancer cells survive in senescence. Blocking the proteins could make chemotherapy more effective. [13]
