# Life Sciences Daily Summary: What Happened on November 7, 2026

**Two cardiovascular drugs miss event-reduction targets despite lowering risk markers**

The day also brought new Zolgensma tumor reports, a melanoma-vaccine filing plan, an FDA cancer approval and a delayed decision for Agios.

## Today in brief

Ziltivekimab and pelacarsen lowered their targeted markers but did not reduce cardiovascular events versus placebo in two trials. [1] [2] Novo Nordisk tested ziltivekimab in ZEUS. Novartis funded HORIZON, which tested pelacarsen against lipoprotein(a), or Lp(a). [1] [2]

Novartis has received nine tumor reports among Zolgensma recipients, including two cases in which it said vector DNA may have entered patients’ genomes. [3] [4] Moderna is preparing an approval filing for its personalized melanoma vaccine with Merck after a Phase 3 trial cut recurrence-or-death risk by 42%. [5] The FDA granted accelerated approval to J&J’s Rybrevant Faspro for certain previously treated advanced head and neck cancers. Agios said the FDA delayed its mitapivat decision in sickle cell disease until February 1, 2027. [6] [7] [8]

## Two cardiovascular drugs lower markers but do not prevent events

In ZEUS, Novo Nordisk’s ziltivekimab lowered interleukin-6, a marker of inflammation, but did not reduce major cardiovascular events versus placebo. [1] Patients taking the drug had no fewer heart attacks, strokes or deaths. Novo stopped two other ziltivekimab trials after a monitoring committee judged them unlikely to succeed. [1]

The Phase 3 HORIZON trial found that pelacarsen substantially reduced Lp(a), a genetically influenced cardiovascular risk factor carried by about one in five people, but did not reduce heart attacks, strokes, urgent coronary procedures or deaths. [2] Participants were followed for nearly five years. Results were presented at the American Heart Association’s scientific sessions and published in the New England Journal of Medicine. [2]

## Zolgensma tumor reports include two possible vector-DNA integrations

Since Zolgensma’s 2019 approval, Novartis has reported nine tumor cases among recipients, seven of them newly disclosed. [3] [4] [9] Novartis said two cases suggest DNA from the therapy’s viral vector entered patients’ genomes. Both patients remain under observation. [3] [4]

Novartis said more than 6,000 patients have received the gene therapy through trials, access programs or routine care. [3] [4] As of November 4, examinations were available for three cases, according to SMA UK and the National Multidisciplinary Team. Two tumors were reportedly associated with treatment, and one was not. [3]

Regulators are reviewing the reports, which cannot establish how often tumors occur or whether Zolgensma caused them. [3] The FDA says adeno-associated virus DNA usually remains outside chromosomes but can sometimes integrate into the genome. The clinical significance of individual integration events is unknown. [3]

## Moderna prepares filing after melanoma-vaccine trial benefit

Moderna said it is preparing to seek approval for intismeran autogene, its personalized cancer vaccine developed with Merck. [5] In the Phase 3 INTerpath-001 trial of 1,137 patients with surgically removed high-risk melanoma, the vaccine plus Keytruda cut recurrence-or-death risk by 42% versus Keytruda alone. [5]

At two years, 82.2% of patients receiving the combination were alive without recurrence, compared with 70.0% of controls. [5] Treatment-related grade 3 to 5 adverse events occurred in 14.2% of combination recipients and 13.4% of controls. Overall-survival follow-up continues. [5]

The vaccine uses each patient’s tumor mutations to select up to 34 targets for T cells. Keytruda blocks an immune-suppressing signal. [5]

## FDA approves Rybrevant Faspro for some advanced head and neck cancers

The FDA granted accelerated approval to J&J’s Rybrevant Faspro for adults whose recurrent or metastatic head and neck squamous cell carcinoma progressed during or after platinum chemotherapy and an immune checkpoint inhibitor. [6] [7] [10] The indication excludes HPV-associated oropharyngeal cancer. It is the drug’s first use outside lung cancer. [7] [10]

In the 102-patient OrigAMI-4 cohort, 42% of patients responded, including 15% with complete responses. Among responders, 56% had responses lasting more than six months. [7] [10] Adverse reactions were serious in 39% of patients and fatal in 6%, including sudden death in 2%. [7] [10]

The FDA based accelerated approval on response rate and response duration from a single-arm study, not a comparison with another treatment. [7] [10] Continued approval may depend on confirmed clinical benefit in Phase 3 OrigAMI-5, which is testing the drug with carboplatin and pembrolizumab as initial treatment. [7] [10]

## Agios cuts R&D workforce as FDA delays mitapivat decision

Agios is cutting more than 35% of its research and development workforce as it shifts resources toward commercial operations and later-stage programs. [8] The company expects about $11 million in restructuring costs this year, savings in 2027 and adjusted profitability in 2028. [8]

The FDA moved its target decision on mitapivat for sickle cell disease from November 1 to February 1, 2027, after requesting more information about a possible safety protocol. [8] According to Agios, the agency is assessing whether the use would need a risk evaluation and mitigation strategy, or REMS. [8]

Mitapivat products generated $55.4 million in global sales in the third quarter, up 24% from the prior quarter. [8] Agios said it is ready to launch and will continue discussing labeling with the FDA during review. [8]

## CMS codifies Medicare negotiation rules for new drug formulations

The Centers for Medicare & Medicaid Services finalized rules covering most existing Medicare drug-price negotiation policies, including how some reformulated products will be grouped. [11] For qualifying fixed combinations, CMS will group formulations when the same application holder adds an ingredient that enables a different route of administration. [11]

The rule applies to drug selection and negotiation in 2027 for prices effective in 2029. It replaces the guidance-based approach after 2028. [11] CMS said grouping certain formulations is intended to prevent manufacturers from shifting patients to a new version to avoid the negotiated maximum fair price. [11]

Commenters argued that developing a new route requires separate clinical, pharmacokinetic, manufacturing and regulatory work, with particular effects on smaller biotech and delivery-technology companies. [11] CMS said the policy covers a narrow category and that it will monitor implementation. [11]

## Armada and Brains agree to combine at about $750 million valuation

Armada Acquisition and Brains Bioceutical signed an agreement to combine, assigning Brains an approximate pre-money equity value of $750 million. [12] The transaction requires regulatory clearances, registration statements and shareholder approval from both companies. Their boards approved it unanimously. [12]

The combined company is expected to be called Brains Therapeutics and list on Nasdaq under the ticker BAPI after closing. [12] The companies scheduled the deal to close in the second quarter of 2027. [12]

## Also worth knowing

- Novo Nordisk will end its research and licensing partnership with Korro Bio on February 1, 2027. The collaboration had been on hold for 12 months and produced no milestone payments for Korro. [13]
- Eli Lilly and Hims & Hers will let patients with prescriptions buy Foundayo from $149 a month and Zepbound from $299 a month through the platform. They said it will not sell compounded versions of Lilly’s GLP-1 drugs. [13]

## The day in numbers

- **9 tumor reports**: Cases Novartis has received among Zolgensma recipients, including two with possible vector-DNA integration. [3] [4]
- **42%**: Reduction in recurrence-or-death risk for Moderna and Merck’s melanoma-vaccine combination versus Keytruda alone. [5]
- **$750 million**: Approximate pre-money equity value assigned to Brains Bioceutical in its planned combination with Armada. [12]
- **February 1, 2027**: New target FDA decision date for mitapivat in sickle cell disease. [8]
