Today in brief

Eli Lilly said retatrutide lowered A1c and body weight more than Ozempic in a head-to-head diabetes study.12 At the highest dose, weight loss was 20.9% with retatrutide and 9.4% with Ozempic among participants who stayed on treatment.1

In Vanda’s Phase 3 trial, fewer participants vomited after taking tradipitant with semaglutide than after taking placebo.3 Idorsia has opened a Phase 3 trial of lucerastat for Fabry disease, focusing its development plan on kidney deposits.4

Retatrutide outperformed Ozempic on weight and A1c

Eli Lilly said retatrutide performed better than Ozempic on blood sugar and weight measures in a head-to-head diabetes study.1 After 80 weeks, A1c fell by 2.2 percentage points with the highest retatrutide dose and by 1.8 points with Ozempic.1

Participants taking the highest retatrutide dose lost 20.9% of their weight, compared with 9.4% for those taking Ozempic.1 Those weight-loss figures include participants who remained on treatment, not the full study population.1

Vanda reports fewer GLP-1-related vomiting episodes with tradipitant

Vanda said its Phase 3 Study 3601 found less vomiting with oral tradipitant than with placebo in people given semaglutide, a GLP-1 receptor agonist.3 Participants received one 1 mg dose of semaglutide, without the usual gradual dose increases, in a trial setup expected to provoke vomiting.3

Vomiting occurred in 43.0% of tradipitant recipients and 58.5% of placebo recipients, a site-adjusted relative reduction of 27%.3 Vomiting with severe nausea occurred in 33.1% and 50.6%, respectively, a 35% adjusted relative decrease.3

Frequent vomiting, defined as more than two episodes on multiple days, affected 6.3% of the tradipitant group and 15.9% of the placebo group.3 Vanda plans to discuss the combined Phase 2 and Phase 3 findings with regulators and submit a supplemental new drug application to the FDA for this use.3

Idorsia starts a new Phase 3 trial of lucerastat in Fabry disease

Idorsia began FAB-KLEAR, a pivotal Phase 3 study of lucerastat in adult men with Fabry disease, shifting its development focus to kidney deposits.4 The company’s earlier MODIFY trial missed its primary endpoint, neuropathic pain, although lower levels of the disease-related substance Gb3 indicated biological activity.4

FAB-KLEAR plans to enroll 16 men and assess kidney biopsy scores after 18 months, with no placebo group.4 Idorsia also plans FAB-SWITCH, a 74-person study that will compare lucerastat with enzyme replacement therapy and assess kidney function.4

Nxera exercised an option to pay 7 million Swiss francs and support global Phase 3 development for exclusive rights in Japan, South Korea, Taiwan, and certain other Asia-Pacific markets, excluding China.4 Idorsia is targeting a possible regulatory filing as early as 2029.4
