Question 9 of 9 Advanced Practical

Design a small-scale model that predicts a critical large-scale bioprocess risk.

Short answer

A useful scale-down model reproduces the large-scale stress relevant to the failure mechanism, then demonstrates that stress predicts the critical response. Matching vessel geometry alone is insufficient.

Why

Large-scale cells move through gradients of substrate, oxygen, pH and CO2 with characteristic exposure and recovery times. A small vessel may be uniformly mixed, so identical average DO or kLa does not reproduce transient cellular experience.

How to work it through

  1. Choose a defined scale-up risk, such as brief oxygen depletion near the feed zone.
  2. Measure or estimate large-scale mixing times, residence-time distributions and gradients.
  3. Construct a two-compartment or controlled-pulse simulator with comparable exposure amplitudes, frequencies and physiological conditions.
  4. Compare growth, metabolites, product titre and quality across scales and deliberately vary stress intensity to test predictive relationship.

An example

A mammalian culture at plant scale shows glycosylation shifts near late-stage high cell density. A scale-down model reproduces the oxygen/CO2 excursions and the direction of the glycan shift across multiple lots, which supports its relevance.

Where the answer stops holding

No single small-scale system reproduces shear, gradients, gas exchange and hydrodynamics simultaneously. A model validated for one cell line or feed regime may not transfer to another.

A strong answer covers

  • A useful scale-down model reproduces the large-scale stress relevant to the failure mechanism, then demonstrates that stress predicts the critical response
  • Matching vessel geometry alone is insufficient
  • Choose a defined scale-up risk, such as brief oxygen depletion near the feed zone

Common mistakes

  • Calling a mini-bioreactor representative because it shares setpoints
  • Overfitting to one failed batch
  • Focusing on titre while ignoring quality attributes
