Question 4 of 9 Intermediate Scenario

How would you distinguish reduced expression from reduced harvest recovery?

Short answer

Separate product created in the bioreactor from product recovered after harvesting. Compare viable biomass, specific productivity and pre-harvest titer with step recovery, mass balances and assay performance. A low downstream output is not automatically a low-expression problem.

Why

Trace the product through the process with a consistent measurement basis. First verify sampling, dilution factors, assay calibration, units and whether the result measures total protein, intact molecule or biological activity. Check cell-growth profiles, viable cell density, metabolites and pre-harvest product titer.

A decrease in titer despite comparable biomass may suggest reduced cell-specific productivity. If pre-harvest titer is normal but post-harvest mass declines, examine lysis, cell-retained product, filter adsorption, clarification losses, hold-time effects and product degradation. Construct a stepwise mass balance that uses collected volume times concentration and documents uncertainty.

In an intracellular product, account for extraction efficiency and relevant compartments. Then look at whether an assay change creates an apparent loss rather than a true one. Proposed process interventions should preserve product quality and follow change-control expectations where required.

Common mistakes

  • Comparing concentrations without accounting for volume and dilution, or equating titer with total recovered mass
