Question 2 of 6 Intermediate Practical

How would you validate an automated sample-preparation workflow?

Short answer

Validate an automated preparation workflow against its intended use with predefined acceptance criteria, representative samples and tests for volume delivery, sample identity, carryover, reproducibility and failure recovery.

Why

Start from the assay requirement and draw the full chain from sample accession to downstream results. Identify critical steps, for example reagent addition, mixing, magnetic-bead washing and barcode transfer, then choose measurable acceptance limits justified by assay performance. Exercise the lowest and highest intended volumes, relevant liquid classes, deck positions, different operators and multiple runs.

Challenge wrong labware, missing tips, scanner errors, pauses and power interruption where feasible. Compare automated and qualified reference workflows using blinded samples and include controls to detect cross-contamination. Record software and firmware versions, method files, consumables and instrument identifiers.

Predefine what constitutes pass, deviation, rework and requalification. A successful demonstration run is not sufficient evidence of routine fitness for use. In regulated contexts, validation depth and approvals depend on intended use and governing requirements.

Common mistakes

  • Relying on a single demonstration plate without predefined limits or exception testing
