Question 19 of 20 Advanced Scenario

What happens after a signal is validated in the EU?

Short answer

A validated signal goes through confirmation, analysis and prioritisation, and assessment, usually by the Pharmacovigilance Risk Assessment Committee (PRAC), ending in a recommendation such as a label change, a study or no action. If it meets the definition of an emerging safety issue, the marketing authorisation holder must notify authorities within 3 working days.

Why

GVP Module IX lists the EU signal management steps: detection, validation, confirmation, analysis and prioritisation, assessment and recommendation for action. Validation asks whether there is enough evidence to justify further analysis, considering the strength of the cases, clinical relevance and previous awareness. Once validated, prioritisation looks at the potential impact on patients and on the benefit-risk balance, which decides urgency.

PRAC assessment may request data from holders, who must cooperate. The outcomes range from updating the product information through a variation, adding a risk to the RMP, requesting a study, to referral procedures for serious concerns. Holders must keep their product information current based on new knowledge and published PRAC recommendations, and report signal work in PSURs.

The faster route is for emerging safety issues, which are concerns urgent and serious enough to need immediate attention. GVP's process figure shows these notified within 3 working days. EMA notes GVP will be updated for Implementing Regulation (EU) 2025/1466, so check current module versions.

A strong answer covers

  • EU signal steps from detection to recommendation
  • Prioritisation by patient and benefit-risk impact
  • PRAC assessment and possible outcomes
  • Emerging safety issue notification within 3 working days
  • Product information updates and PSUR reporting

Common mistakes

  • Treating a validated signal as a confirmed risk.
  • Waiting for the next PSUR to report an urgent safety issue.
  • Assuming the holder has no role once PRAC takes over.
