Question 20 of 20 Advanced Scenario

Does moving an operating point within an approved design space need revalidation?

Short answer

From a regulatory filing view, working within an approved design space is not a change, so no variation or supplement is needed. From a quality system view, the move still goes through change control, which judges whether scale, equipment or monitoring evidence supports it and whether verification batches are prudent.

Why

ICH Q8(R2) defines a design space as the combination of input variables and process parameters demonstrated to assure quality, and says working within it is not considered a change, while moving outside is a change that normally starts a regulatory post-approval process. ICH Q10 adds the important second half: from a pharmaceutical quality system standpoint, all changes should be evaluated by the change management system. So the regulatory question and the quality question get different answers.

The quality evaluation asks how the design space was established. Q8(R2) notes that a design space can be developed at any scale, but the applicant should justify its relevance to production scale and discuss scale-up risks. If the edge of the design space was mapped only at small scale, moving the commercial set point there may carry more risk than the filing suggests.

The evaluation also asks whether interactions were studied, whether the move combines with other parameters already near their edges, and whether monitoring will detect a problem. For example, raising granulation water to the upper edge of a design space studied at 10 kg, on a 300 kg commercial line, might justify a few closely monitored verification batches and heightened CPV sampling even though no filing is needed. A company that skips change control on the grounds that the design space allows it will struggle to explain the decision when something goes wrong.

A strong answer covers

  • Not a regulatory change within design space per Q8(R2)
  • Still evaluated through change control per Q10
  • Scale relevance of the design space
  • Interactions and combined edges
  • Verification batches and heightened monitoring as prudent options

Common mistakes

  • Skipping change control because the move is within the design space.
  • Assuming a design space mapped at small scale is equally reliable at its edges at commercial scale.
  • Treating a proven acceptable range for one parameter as a multivariate design space.
