Question 4 of 6 Intermediate Concept

Why is barcode traceability crucial in automated sample preparation?

Short answer

Barcode traceability preserves the link between a physical sample, its location, each automated transfer and the final result. A correct measurement is still unusable if its identity is wrong.

Why

Picture a 96-well plate moved to a robot after accessioning. A plate rotation, misread barcode or incorrect source-to-destination map can silently assign a valid result to another person or experiment. Use stable identifiers and validated scanner reads at critical custody changes, record plate and well positions, enforce expected container states and reject mismatches before dispensing.

Map parent samples to aliquots with versioned transfer events that include instrument, run, operator and timestamps. Include controls for duplicate labels, damaged codes and human overrides. Reconcile counts and identities between accession, preparation and result import, retaining an audit trail when exceptions are investigated.

Barcoding alone does not prove correctness if the barcode was attached to the wrong tube, so link physical process controls with software constraints.
