Question 5 of 6 Advanced Scenario

How does lifecycle management affect an already approved product?

Short answer

An approval establishes a controlled baseline, not permission to change everything freely. Changes require documented assessment of quality, safety, efficacy and region-specific reporting or approval obligations.

Why

If a biologics manufacturer changes a chromatography resin, assess potential effects on product attributes, process validation and analytical comparability. Internal change control should document risk, supporting experiments, manufacturing history and affected filings. Determine whether the change alters established conditions or other regulatory commitments, and choose the correct notification or approval pathway in each market before implementation where required.

ICH Q12 offers a common framework for managing postapproval CMC changes, but regional legal implementation differs. The EU revised variation regulation applied from January 2025 and the updated guidelines from January 2026. Specific procedures still depend on product and change type.

Monitor changes after implementation, maintain dossier consistency and update product information when appropriate. Successful internal verification alone does not necessarily satisfy external regulatory obligations.
